So we noticed something very similar when we 3D printed scaffolds from plant based materials and seeded them with fibroblasts....is that the cells were most aligned and viable around the edges and ridges. One of the reasons this could be is that ECM mechanical properties play a crucial role in determining cell orientation and so mechanical properties on the ridges are highest hence fibroblasts will more likely align and grow there most. As a former biomedical engineer who studied tissue engineering and biomaterials, I really like how you simplified the communication surrounding what each of the compounds you were using were for. HBSS, DMEM, FBS... It's amazing what academics take for granted with knowledge of what they think are "basic" things that are quite literally so fundamentally important.
